Application: Indicator for the microbiological assessment of the effectiveness of high-pressure steam sterilisation. Biological test. A filter paper strip impregnated with a suspension of spores (of the strain Bacillus stearothermophilus ATCC 7953) capable of reverting to vegetative forms. The indicator complies with the requirements of the European Pharmacopoeia. D-value for a temperature of 121°C for approx. 2.5 minutes, determined using the survival curve method (the time required to reduce the number of viable microorganisms to 10% of the initial count). Place the test indicators in at least two of the largest packages representative of the given batch and position them in the areas least accessible to the sterilising agent. Once the sterilisation process is complete, transfer the discs under aseptic conditions to test tubes each containing 10 ml of casein-soya broth. Incubate at 56 °C for 7 days. If sterilisation has been ineffective, the broth will become cloudy and form a scum. If sterilisation was successful, the broth remains clear. At the same time, a culture of an indicator not subjected to the sterilisation process should be carried out. Growth in this test tube should occur within 48 hours at the latest.
